I'm trying to set my site up so that orders over $35 must go out UPS (any UPS method) instead of USPS. Is there any way to do this?

Yes, just edit the Workflow-ng conditions for the UPS and USPS shipping methods, so that UPS is only applied when Order Total > $35, and USPS is only applied when Order Total <= $35.
