Dear Support,

I have a website started for a friend at www.mydomain.com.au - they want to have sales.mydomain.com.au

I have installed drupal again but now realise, it's going to be a completely different site.
Should I be using a drupal multi-site install instead to make my life easier?

From reading this topic http://www.ubercart.org/forum/support/1634/individual_stores it appears so.

Could someone confirm and give any hints the easiest way to do this?

Take a look at http://drupal.org/project/subdomain subdomain module. It is simple to install and configure, could be a more accurate solution to your task.
